The blockade is the latest blow to efforts to implement large-scale vaccination programs against a deadly virus that has killed more than 2.7 million people worldwide and damaged the global economy.
"As a precaution, vaccinations should be suspended during the current investigation period," the Hong Kong government said in a statement.
Both Chinese cities said they made the decision after being contacted by Chinese pharmaceutical company Fosun about the issue while distributing Pfizer / biotech vaccines in China.
Defective packaging has been found in 210,102 laths, officials said.
Statements from Hong Kong and Macau did not elaborate on how bad the packaging was, but both said they did not believe there were safety issues.
